BIH Core Facility Proteomics

Investigating living systems at the protein level is one of the most powerful ways to provide important insights into many biological processes across all kingdoms of life. Studying proteins can unravel new functions, and whole layers of regulation exist that are not accessible with DNA- or RNA-based techniques, in particular by degradation or post-translational modification.

Mass spectrometry-based proteomics offers the possibility of measuring thousands of proteins in a single run and has fundamentally changed the way that biological systems are analyzed. Liquid chromatography coupled mass spectrometry has reduced the time necessary for the determination of a proteome to a couple of days, and, recent advances in quantitative proteomics allow the development of sophisticated models based on the high quality, quantitative measurements achieved using isotopically-coded amino acids in both metabolic labeling techniques (SILAC) and  spike-in reference peptides used in targeted proteomic approaches (AQUA). These tools enable the precise determination of biological responses to stimuli or general differences between cells and tissues.

Analysis methods

The BIH Core Facility Proteomics will offer different shotgun measurement techniques for the determination of deep proteomic data sets. In addition to the large-scale deep analysis, several techniques for the identification of interacting proteins or post-translational modifications will be available, as well as techniques for the targeted quantification of specific proteins can be set up. The exact nature of the measurement will be tailored to the experimental question posed by the research group.
