Guest Lecture at the BIH Center for Regenerative Therapies (BCRT)

Epigenetic mechanisms play a crucial role in establishing and preserving cellular states and function throughout an organism’s life. DNA methylation is an essential part of this multilayered regulation and displays a highly conserved, characteristic bimodal distribution across most somatic cell types. However, in tumors, the distribution of DNA methylation is fundamentally different, making DNA methylation an ideal readout for tumor-type prediction from sequencing data. Specifically in the field of neuropathology, DNA methylation-based tumor type classification has recently become part of the WHO. However, standard practical care relies on a multi-day procedure with long waiting times for clinicians and patients. Recent advances in third-generation sequencing technologies have brought the chance of intraoperative classification into reach. To enable CNS tumor type classification within less than one hour, we developed a new framework enabling live classification of CNS epigenomes, which can run parallel to an ongoing Nanopore experiment, resulting in classification results within 15 minutes of sequencing.

About the Speaker

Helene Kretzmer is a leading expert in computational genomics. Since October 2024, she has held a W2 professorship and chair at the Hasso Plattner Institute, University of Potsdam. She also leads a bioinformatics research group at the Max Planck Institute for Molecular Genetics (MPIMG) since 2022. Her scientific focus lies in genomic regulation and computational analysis of sequencing data, particularly in cancer genomics. She completed her PhD in Informatics in 2016 at the University of Greifswald, specializing in DNA methylation analysis.
